Padrón Cigars was founded on September 8, 1964, in Miami, Florida, by Cuban-born José Orlando Padrón. The Padrón family had grown tobacco for generations in the Cuban province of Pinar del Río, with ancestors who had farmed tobacco in the Canary Islands before emigrating to Cuba. After leaving Cuba following the revolution, José Orlando Padrón saved enough money to rent a storefront in Little Havana and, with a partner, began producing 200 hand-rolled cigars per day.

In 1970, Padrón moved the company's manufacturing to Estelí, Nicaragua, beginning the relationship with Nicaraguan tobacco that would define the brand. The new rolling center grew over decades into 17 buildings owned by Padrón in Estelí, with usable space totaling 75,000 square feet, mostly dedicated to tobacco storage and aging. The 1964 Anniversary Series was released in 1994 to celebrate 30 years in business. The 1926 Series followed in 2002, marking the 75th birthday of José Orlando Padrón.

Both lines became regular winners of "Cigar of the Year" honors from Cigar Aficionado. José Orlando Padrón passed away in 2017 at age 91. The company remains family-owned and is currently operated by his sons Jorge and Orlando Padrón. Padrón cigars are sold only through Padrón's established appointed merchant network, with strict pricing and distribution controls intended to protect retailer margins.
